    Evaluation of interspecific DNA variability in poplars using AFLP and SSR markers

    The objective of this paper was to examine interspecific DNA variation in poplars using AFLP and SSR markers. The AFLP and SSR markers polymorphism and its power of discrimination were determinedwithin 13 genotypes of different genetic background (clones, cultivars, hybrids) of two sections (Aigeiros and Tacamahaca) of genus Populus. Twelve sets of PTR and four sets of WPMS microsatellite primers as well as 6 AFLP primer combinations were used in this study. SSR and AFLP markers revealed high power of discrimination, 0.81 and 0.92 respectively. Results obtained using SSR data showed a clear separation of three major clusters, while four major clusters were obtained using AFLP data. Both markers crearly separated two distinct clusters, one included Populus nigra and the other Populus deltoides genotypes. According to both markers, different genetic background were revealed between two clones of Populus trichocarpa. When evaluating polymorphisms within genotypes ofinterest, microsatellite and AFLP DNA markers proved to be a useful tool for distinguishing genetic background of tree clones, cultivars and hybrid genotypes thus grouping them according to their genetic dissimilarity. The results presented in this study could be of significant interest in poplarbreeding programs and could also be used as a valuable annex in new bred clones registration process

    Screening of drought oxidative stress tolerance in Serbian melliferous plant species

    This study was designed to examine and compare antioxidant and free-radical scavenging activities of leaves of six different melliferous plant species (Populus alba, Robinia pseudoacacia, Sophora japonica, Euodia hupehensis, Tilia sp., Fraxinus sp.) from Serbia in order to evaluate their drought oxidative stress tolerance. Experiment was conducted during June, July and August. In this study, we reported the results concerning proline accumulation, soluble protein content, quantities of malonyldialdehyde,total antioxidant capacity determined by FRAP method and scavenger activity determined by DPPH method. According to our results, all melliferous plant species were subjected to drought oxidative stress during July when soil humidity decreased. During July, proline content and MDA quantity increased and soluble proteins decreased in all investigated species. High and permanent antioxidant activity during the whole investigated period was observed in P. alba, but insufficient to protect itsleaves from oxidative injury during the period of drought in July. The highest ability to accumulate proline and highest protein content under severe drought stress in July was observed in Fraxinus sp. Other investigated antioxidant parameters (total antioxidant and DPPH radical scavenger capacities) were high and accumulation of MDA was low which indicate high drought oxidative stress tolerance. Therefore, highest ability to adapt under severe drought stress and highest drought oxidative stresstolerance were observed in Fraxinus sp.Key words: Melliferous trees, lipid peroxidation, DPPH, FRAP, proline accumulation

    Physical and mechanical characteristics of Hisex Brown hen eggs from three different housing systems

    The aim of this study was to compare physical and mechanical characteristics of Hisex hen eggs collected from three different housing systems: enriched cage housing, aviary housing, and free-range systems. The following physical and mechanical characteristics if eggs were compared: dimensions, surface area, volume, sphericity, shape index, shell thickness, weight, composition, yolk to albumen ratio, rupture force, specific deformation, absorbed energy, and firmness. The largest and heaviest eggs were collected from cage housing, followed by eggs from free-range systems and aviary housing. According to shape index, eggs from aviary housing can be described as round, while eggs from cage housing and free-range systems can be characterised as normal or standard. Eggs from free-range laying hens had the highest yolk percentage and yolk to albumen ratio (26.2% and 0.427). In comparison to eggs from aviary housing and free-range systems, eggs from enriched cage housing had the thickest shells and the highest shell strength, and required the highest force to rupture those eggs. The average force required to rupture Hisex Brown hen eggs from cage housing in all three axes was 44.14 N, which was 12.1% higher than the average force required to rupture eggs from a free-range system (39.37 N) and 17.1% higher than the average force required to rupture eggs from aviary housing (37.68 N). The highest forces required to rupture eggs from all three housing systems were determined on loading along the X-front axis and the lowest forces were determined along the Z-axis. The results obtained in this study can be useful to producers when selecting hen housing systems in order to reduce egg damage during storage and transport.Keywords: egg composition, egg weight, rupture force, shape index, shell thicknes

    Comparative Study of Hematopoietic Differentiation between Human Embryonic Stem Cell Lines

    Directed differentiation of human embryonic stem cells (hESCs) into any desired cell type has been hailed as a therapeutic promise to cure many human diseases. However, substantial roadblocks still exist for in vitro differentiation of hESCs into distinct cell types, including T lymphocytes. Here we examined the hematopoietic differentiation potential of six different hESC lines. We compare their ability to develop into CD34+ or CD34+CD45+ hematopoietic precursor populations under several differentiation conditions. Comparison of lymphoid potential of hESC derived- and fetal tissue derived-hematopoietic precursors was also made. We found diverse hematopoietic potential between hESC lines depending on the culture or passage conditions. In contrast to fetal-derived hematopoietic precursors, none of the CD34+ precursors differentiated from hESCs were able to develop further into T cells. These data underscore the difficulties in the current strategy of hESC forward differentiation and highlight distinct differences between CD34+ hematopoietic precursors generated in vitro versus in vivo

    Notch signaling is critical during multiple stages of T cell development in both mouse and human. Evidence has emerged in recent years that this pathway might regulate T-lineage differentiation differently between both species. Here, we review our current understanding of how Notch signaling is activated and used during human T cell development. First, we set the stage by describing the developmental steps that make up human T cell development before describing the expression profiles of Notch receptors, ligands, and target genes during this process. To delineate stage-specific roles for Notch signaling during human T cell development, we subsequently try to interpret the functional Notch studies that have been performed in light of these expression profiles and compare this to its suggested role in the mouse

    Antitumor activity from antigen-specific CD8 T cells generated in vivo from genetically engineered human hematopoietic stem cells

    The goal of cancer immunotherapy is the generation of an effective, stable, and self-renewing antitumor T-cell population. One such approach involves the use of high-affinity cancer-specific T-cell receptors in gene-therapy protocols. Here, we present the generation of functional tumor-specific human T cells in vivo from genetically modified human hematopoietic stem cells (hHSC) using a human/mouse chimera model. Transduced hHSC expressing an HLA-A*0201–restricted melanoma-specific T-cell receptor were introduced into humanized mice, resulting in the generation of a sizeable melanoma-specific naïve CD8^+ T-cell population. Following tumor challenge, these transgenic CD8^+ T cells, in the absence of additional manipulation, limited and cleared human melanoma tumors in vivo. Furthermore, the genetically enhanced T cells underwent proper thymic selection, because we did not observe any responses against non–HLA-matched tumors, and no killing of any kind occurred in the absence of a human thymus. Finally, the transduced hHSC established long-term bone marrow engraftment. These studies present a potential therapeutic approach and an important tool to understand better and to optimize the human immune response to melanoma and, potentially, to other types of cancer

    Positive airway pressure (PAP) treatment reduces glycated hemoglobin (HbA1c) levels in obstructive sleep apnea patients with concomitant weight loss: Longitudinal data from the ESADA

    Patients with obstructive sleep apnea (OSA) are at increased risk of developing metabolic disease such as diabetes. The effects of positive airway pressure on glycemic control are contradictory. We therefore evaluated the change in glycated hemoglobin (HbA1c) in a large cohort of OSA patients after long-term treatment with positive airway pressure. HbA1c levels were assessed in a subsample of the European Sleep Apnea Database [n=1608] at baseline and at long-term follow up with positive airway pressure therapy (mean 378.9±423.0 days). In a regression analysis, treatment response was controlled for important confounders. Overall, HbA1c decreased from 5.98±1.01% to 5.93±0.98% (p=0.001). Patient subgroups with a more pronounced HbA1c response included patients with diabetes (−0.15±1.02, p=0.019), those with severe OSA baseline (−0.10±0.68, p=0.005), those with morbid obesity (−0.20±0.81, p<0.001). The strongest HbA1c reduction was observed in patients with a concomitant weight reduction >5 kilos (−0.38±0.99, p<0.001). In robust regression analysis, severe OSA (p=0.038) and morbid obesity (p=0.005) at baseline, and weight reduction >5 kilos (p<0.001) during follow up were independently associated with a reduction of HbA1c following PAP treatment. In contrast, PAP treatment alone without weight reduction was not associated with significant Hb1Ac reduction. In conclusion, positive airway pressure therapy is associated with HbA1c reduction in patients with severe OSA, in morbidly obese patients. and most obviously in those with significant weight lost during the follow-up. Our study underlines the importance to combine positive airway pressure use with adjustments in lifestyle to substantially modify metabolic complications in OSA
